Hayne, Knights skipper Kurt Gidley and Penrith forward Luke Lewis will be the three major changes for Origin II in Sydney.
After controversially being overlooked for Origin I, Hayne will be welcomed back into the fold when coach Ricky Stuart names his squad on Sunday night.
Hayne will be named on the wing, replacing St George Illawarra flyer Brett Morris, who injured his hamstring last weekend.
Hayne admitted that selection for the Blues was weighing on his mind.

"It's obviously something I think about, but something I try not to worry about. So if I get selected, I get selected," Hayne said.
While Hayne's return will dominate the headlines, the Blues squad for their must-win clash at Homebush on June 15 will also feature three significant changes.
Gidley is back to add experience, versatility and spark from the bench.
Morris' Dragons teammate Dean Young is likely to may way for Gidley - a tough decision for Stuart, who is a huge supporter of the utility forward.
Cronulla's Test prop Kade Snowden is also expected to be a casualty from Origin I in Brisbane because back and shoulder problems have affected his form in recent weeks.
Snowden and front-row teammate Jason King came under heavy criticism for their efforts in game I.
King is expected to survive the axe, but he needed a strong performance against the Bulldogs oin Saturday night.
It is anticipated that Eels prop Tim Mannah will be elevated to a starting role.
Mannah was strong from the bench in Brisbane and would play alongside King.
He suffered a minor rib injury in the Eels' 14-all draw with the Dragons on Friday night, but he assured Blues officials he was "sweet".
King's Sea Eagles teammate Anthony Watmough is also a chance to be included in the squad, despite claims earlier this season that a string of off-field misdemeanours had thwarted his representative aspirations.
But Watmough's form for Manly is too good to ignore.
It's just a matter of how Stuart can fit the former Test back-rower into his side.
The win-at-all-costs attitude from the Blues will also see Penrith strike weapon Lewis named on the bench.
The Australia representative and Panthers leader was unavailable for selection before Origin I because of injury, but regular phone conversations with Stuart over the past month inspired the 27-year-old to ensure his availability for game II.
Lewis proved his worth with a man-of-the-match display against South Sydney last Monday night.
Canberra and Test prop Tom Learoyd-Lahrs will have one final chance to impress Blues officials when the Raiders host North Queensland Cowboys this afternoon.
But it appears even a match-winning performance won't be enough for the hulking prop. Concerns over his fitness levels are a contributing factor to his fall from grace this season.
Jamie Soward and Mitchell Pearce will retain the job of steering the Blues around the park, while Bulldogs hooker Michael Ennis will wear the No.9 jumper after coming off the bench in Brisbane.
Beau Scott, Greg Bird and Paul Gallen will form the back row.
In making just three changes, Stuart has delivered on his promise to show faith in the side that narrowly went down 16-12 at Suncorp Stadium a fortnight ago.
Gallen said that he was eagerly anticipating a sellout crowd at Homebush for the do-or-die encounter.
"We'll be much better as a unit for the experience of Origin I and I'm really looking forward to getting back and playing in front of our home fans," Gallen said.
"We came together really well as a group ahead of game one and I think we'll only kick on from where we left each other. I know we've got the team to do NSW proud."
